Enzymes are biocatalysts produced by living cells to bring about specific biochemical reactions generally forming parts of the metabolic processes of the cells. Enzymes are highly specific in their action on substrates. Enzyme lipase (triacylglycerol acylhydrolase) is a unique enzyme which can catalyze various types of reactions such as hydrolysis, esterification, alcoholysis etc. In particular, hydrolysis of vegetable oil with lipase as a catalyst is widely studied. Today, the immobilization procedure shows a great potential for commercial applications of the immobilized lipase, a relatively low cost commercial enzyme. This paper presents the result of studying on producing fixed lipase enzyme on microparticle “chitosan-Fe

O
4
”, microparticle is a complex of nano particles Fe
3
O
4
being absorbed on chitosan so it has magnetic property, enzyme links to microparticle through an intermediate brigde – glutaraldehyde. Free enzyme which is used to fix is commercical lipase enzyme of Sigma (Life science) being extracted from pancreas of pig. After producing fixed lipase enzyme, determining its optimum conditions: pH = 6.0, t = 40

C and enzyme activity 185 IU/mg correspoding to optimum pH, temperature and 3 hours of reaction time; the productivity of attaching lipase enzyme to the carrier achives 75.1%; the reaction could be stopped and recover enzyme by the magnetic force; the enzyme could be reused many times. Further research will use manufactured fixed lipase to carry out este transposition reaction in biodiesel production from vegetable oil and animal fat.
